Antiserum to bovine thyroglobulin was conjugated with fluorescein isothiocyanate at coupling ratios of 5 mg and 2.5mg per 2ml of the antiserum, and fractionated by means of DEAE-cellulose column chromatography. Each fraction was analyzed for its absorption values at 280mμ, and 495mμ and antibody titer, and compared with each other.<BR>The conjugate of a coupling ratio of 2.5 mg per 2ml is superior to that of 5 mg, and the concentration of NaCl in eluting solution ranging from 0.01 to 0.025 M seems to be suitable. But, fluorescence of freeze-substituted sections stained with these fractions is weak and does not permit the evaluation of this method superior to Marshall's method.
