Tentative Preparation of an Inactivated Virus Vaccine for Equine Viral Arteritis

An attempt was made to develop an inactivated virus vaccine for equine viral arteritis (EVA) which might be used safely in the areas free from the disease. The Bucyrus strain of EVA virus was propagated at moi (multiplicity of infection) 0.6 in roller bottle cultures (840cm2 in growth area) of E. Derm cells maintained in 25ml of serum-free medium 199 containing 0.2% horse albumin. The precipitation of culture fluid with 8-10% polyethylene glycol was successfully used to concentrate virus antigen and reduce extraneous protein substance. No infectious virus was detectable 6h after treatment with 1:1000 (0.1%) formalin at 37°C. When inoculated experimentally with virus antigen inactivated with formalin for 48h, 4 horses appeared to be safe, showing no side effects clinically. They produced high titer of serum-neutralizing antibody after the second inoculation. The titer was much higher when this inoculation was done 4 weeks than 2 weeks after the first inoculation.
