Immunoelectron microscopic study on the localization of secretory component in lung by the post-embedding method.

The localization of secretory component (SC) in normal bronchial cells and pulmonary adenocarcinoma cells was studied immuno-electronmicroscopically by the postembedding method using the low temperature embedding procedure.<BR>SC was localized in perinuclear spaces, endoplasmic reticulum, Golgi complexes, plasma membrane and occasionally cytoplasmic vesicles of the bronchial mucous and serous gland cells. SC was also present in mucous and serous granules. In ciliated bronchial epithelial cells, SC was only demonstrated in the supranuclear region<BR>It was also found that SC was synthesized by adenocarcinoma cells in the area of the endoplasmic reticulum and Golgi complexes and was present on the plasma membrane. However, the difference of the polarity of SC on the plasma membrane between normal and malignant cells was not detected.<BR>These findings were almost same as those previously obtained by the pre-embedding method except for the polarity on plasma membrane. It was suggested that this postembedding method is a useful technique to immunohistochemically detect glycoprotein in the cell matrix and organelles.
