Callus induction and plant regeneration from seed and shoot tip explants of Zoysia minima collected from New Zealand

This study investigated the effects of a basic medium supplemented with concentrations of 2, 4-D, silver nitrate (AgNO<SUB>3</SUB>), carbohydrate source and BA application on callus formation and plant regeneration from the seeds and shoot tips of <I>Zoysia minima</I> collected from New Zealand. The high regeneration frequency from seeds was observed from calli initiated on LS basal medium containing relatively low concentration (0.5 and 1 mg/l) of 2, 4-D. No stimulatory effect of AgNO<SUB>3</SUB> was observed on compact callus induction and plant regeneration. As a result of comparing 5 sources of carbohydrate, maltose was a comparatively better carbohydrate that was supplemented with the regeneration medium. It appears that compact and friable ca11i were initiated at the base of the shoot tips inoculated approximately 10 days after culture initiation. The highest regeneration frequency from shoot tips was observed when the compact calli induced on the medium with 1mg/1 2, 4-D were transferred to the regeneration medium with relatively lower concentration (0.5-2.0 mg/l) of BA . In this experiment, a successful regeneration method for compact callus derived from shoot tips was developed and shoot tips were found to be a highly regenerative explant source for <I>Z. minima</I>.
