Effect of Tetracycline Hydrochloride (TC-HCl) Coating to Nucleus on Pearl Oyster (Pinctada fucata martensi) Survival after Nucleus Insertion.

In order to improve the survival rate of pearl oyster after nucleus insertion, pearl nuclei were immersed in a coating solution containing 0.2% tetracycline hydrochloride (TC-HC<I>l</I>) and 0.4% succinated atherocollagen, followed by coating with polyethylene glycol 6000 to keep surface dry. Each nucleus was coated with 200-220μg of TC-HC<I>l</I>. The TC-HC<I>l</I>-coated nucleus formed an anti-bacterial circle against the following bacteria strains: <I>Edwardsiella tarda</I> NuF84, <I>Enterococcus seriolicida</I> NG8206, <I>Vibrio</I> sp. KP-01, <I>V. anguillarum</I> and <I>Pseudomonas fluorescens</I>. After the insertion, TC-HC<I>l</I> was gradually absorbed into the shellfish tissues. Their TC-HC<I>l</I> content reached the highest level of 133μg/individual 6 hours after the insertion, and then started to decreased, reaching 10μg/individual on the 7th day. Tissue concentration of TC-HC<I>l</I> was high in the visceral mass compared to that in the mantle, gill, and adductor muscle. To ascertain the safety of TC-HC<I>l</I> to the shellfish itself, nuclei coated with four times higher concentration of the antibiotic were inserted to pearl oysters and the oysters were kept for 45 days. The survival rate of the antibiotic treated group (86.7%) was higher than that of the control group (63.3%; inserted with untreated nuclei) . It was almost the same as that of the untreated group (83.3%) . The effect of TC-HC<I>l</I> coating was also examined with large scale field experiments in Kochi and Ehime Prefectures. In the field conditions, the survival rates of the treated groups were higher than those of the control groups. The nuclei retaining rates were also higher in the treated group.
