Development, Growth, and Survival Rate of Protoplasts and Sporelings of Laver (Porphyra yezoensis) in Various Culture Media.

The present work was carried out to find suitable medium for the growth of protoplasts and sporelings of layer, <I>Porphyra yezoensis</I>. Eleven media including variants of enriched seawater (ESS, modified ESS, PES, modified SWM III, F/2, SWII, and modified ESP) and artificial seawater (ASP<SUB>12</SUB>NTA, modified ASP6, ASS<SUB>1</SUB>, and Jamarin U) were tested in comparison with natural seawater (Salinity 32.5 ‰) .<BR>The results obtained are summarized as follows, <BR>1) The survival rates of protoplasts in ASS<SUB>1</SUB> and modified ASP6 were inferior to that in natural seawater after 19 days.<BR>2) In enriched seawater media (PES, modified SWMIII, ESS, and F/2) and artificial seawater media (ASP<SUB>12</SUB>NTA and Jamarin U), over 60 % of protoplasts regenerated to monospore-produicing form against 38 % in natural seawater. Only 6 % of protoplasts in ASS<SUB>1</SUB>, regenerated to it form within 19 days. No regeneration was observed in modified ASP6.<BR>3) The size of monospore-producing, callus-like, and rhizoidal form in modified SWMIII, ESS, and ASP<SUB>12</SUB>NTA were larger than those in the other media after 19 days.<BR>4) The growth of the sporelings was promoted in modified SWMIII, ESS, ASS<SUB>1</SUB>, and ASP<SUB>12</SUB>NTA.<BR>5) These results indicate that modified SWMIII, ESS, and ASP<SUB>12</SUB>NTA are suitable media for culture of protoplasts and sporelings of <I>P. yezoensis</I>.
