Preparation of Euglena gracilis Enriched with Ecosapentaenoic and Docosahexaenoic Acids.

Incorporation and accumulation of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in <I>Euglena gracilis</I> were investigated for its application as feed for living feed of fish larvae and fry after enriching with the n-3 highly unsaturated fatty acids (n-3 HUFA) . While supplementation of pollack or cuttlefish liver oil as a source of EPA and DHA resulted in little incorporation of these acids in <I>E. gracilis</I>, a mixture of free fatty acids obtained by hydrolysis of cuttlefish liver oil caused marked incorporation of them in the cells. When crude EPA or DHA was added to the growing medium, the contents of both lipid and these fatty acids in the total fatty acids in <I>E. gracilis</I> cells were increased. The more supplementation of EPA or DHA gave higher content of these acids in the total fatty acids in <I>E. gracilis</I>. In this way, <I>E. gracilis</I> cells containing more than 75% of EPA or DHA in the total fatty acids were obtained by the supplementation of purified EPA or DHA. Supplementation of ethyl esters of these acids resulted in less increases of the lipid content and the contents of these acids in the total fatty acids.<BR>Results of these experiments show that the contents of EPA and DHA can be increased extensively in <I>E. gracilis</I> by supplementing these acids into growing media, and the harvested cells are expected to be profitable in use as an enriched feed planktons, living feed for fish larvae and fry.
