Differences in Chemical Composition and Antigenicity between Two Ribosomal Vaccines Prepared from Pasteurella piscicida.

It was reported that protection in yellowtail, <I>Seriola quinqueradiata</I> induced by ribosomal precipitate fraction (RBP) was significantly higher than that by ribosomal supernate fraction (RBS) . In this study, chemical composition and antigenicity of RBP were investigated and compared with those of RBS.<BR>RBP and RBS were prepared from <I>Pasteurella piscicida</I> by the method of Ninomiya <I>et al</I>. Ribonucleic acid (RNA), protein and lipopolysaccharide (LPS) content of RBP and RBS were also measured. Furthermore, antigenicity of RBP and RBS were investigated by immunodiffusion assay using anti-RBP and anti-RBS rabbit serum.<BR>The major component of RBP and RBS was RNA and protein respectively, although both contained a little amount of LPS. RBP and RBS formed continuous precipitation lines on immunodiffusion plate when anti-RBP or anti-RBS rabbit serum were used. Anti-RBP rabbit serum reacted with RBP, RBS, LPS and purified LPS, whereas anti-RBS rabbit serum reacted only with RBP and RBS on immunodiffusion assay. These results suggested that LPS contained in RBP which is responsible for the antigenicity is different from that in RBS.
