Characteristics of two virus strains isolated from young Japanese flounder Paralichthys olivaceus.

Two diseases with different symptoms were observed among cultured young Japanese flounder <I>Paralichthys olivaceus</I> in 1986 and 1987. One was characterized by development of ascites and the other by haemorrhage in the head. To investigate the cause of the diseases, virological examination was carried out. Virus was isolated from Japanese flounder of both diseases using RTG-2 or CHSE-214 cells. Intraperitoneal injections of the two virus strains to healthy young Japanese flounder resulted in ascites. The virus could be reisolated from the fish. Electron microscopy revealed unenveloped hexagonal particles about 63 nm in diameter in CHSE-214 cells. Replication of both virus strains were not affected in cell cultures by 50 μ<I>g</I> iododeoxyuridine. Both strains were stable against chloroform, at pH 3, and 56 °C for 30 min. Both strains were neutralized by anti-IPNV Ab rabbit serum. Both strains replicated in CHSE-214, RTG-2, FHM, EPC, BF-2 and EK-1 cells and at temperature range of 10-30°C. From these results, it was concluded these two virus strains are pathogenic to young Japanese flounder. The virus strains are thought to be Birnaviruses.
