Production of transgenic mouse carrying human luteinizing hormone .BETA. subunit gene under the control of rabbit .BETA.-casein promoter.

To examine the tissue-specific expression of the transgene <I>in vivo</I>, we have generated a transgenic mouse lineage producing transgene products in mammary gland. The gene was constructed by fusing a 2.1 Kb fragment of the rabbit β-casein gene 5'flanking region to a 1.4 Kb genomic fragment of the human luteinizing hormone β subunit gene (rβ-casein/hLHβ) . Four transgenic mice were generated and secreted hLH β subunit into milk at the concentration between 0.17 and 0.21 ng/m<I>l</I>. Unexpectedly, the foreign protein was also detected in serum, and the mRNA of the rβ-casein/hLHβ transgene was expressed in some other tissues including ovary, uterus, testis, thymus, salivary gland and brain. In addition, the transcription of the endogenous β-casein gene was observed in the same tissues but not in the brain. These findings suggest that <I>de novo</I> expression of the β-casein gene in normal mice is not tissue-specific to mammary gland and that the 5'flanking region of rabbit β-casein constructed in this experiment possibly promotes a foreign protein in several tissues besides mammary gland in transgenic mice.
