The extract obtained from phagocytosing neutrophils enhanced the phagocytic activity of neutrophils. The generation of phagocytosis-stimulating factor (PSF) by neutrophils reached the maximum at 60 min after the start of phagocytosis. The PSF activity was found in a Sephadex G-100 fraction containing molecular weight of approximately 18, 000 daltons. PSF was stable to heating at 80°C for 30 min but destroyed completely at 100°C for 30 min. PSF was inactivated by tryptic or chymotryptic digestion, indicating its protein nature. PSF was present in the granular fraction of phagocytosing neutrophils, and the generation of this factor was inhibited by cytochalasin B and colchicine, but not by cycloheximide, suggesting that the generation of PSF by neutrophils would result from the activation of PSF-precursor in the granules through the fusion of phagosomes with lysosomes.
