<I>Effect of CCA on the experimental amyloidosis induced by lipopolysaccharide in C3h/He mice</I>

CCA [N- (2-carboxyphenyl) -4-Chloroanthranilic acid disodium salt] is a new antiarthritic and immunomodulating compound synthesized and developed by Chugai Pharmaceutical Co., Ltd.. CCA was effective to prolong the life-span of female (NZB × NZW) F<SUB>1</SUB> mice and to inhibit the secondary inflammation in adjuvant arthritic rat while CCA was totally ineffective against carrageenin-edema and cotton pellet granulomatosis in rats.<BR>In this article, it was also shown that CCA (50 mg/kg×7 days, p.o.) inhibits thymic atrophy of mice caused by cortisone administration. Similar effect of CCA was observed in alcohol-induced thymic atrophy of rats. CCA enhanced spleen cell responses to PHA in normal mice.<BR>From the aspects of these results, effect of CCA on experimental murine amyloidosis was investigated. Interestingly, CCA delayed the induction of secondary amyloidosis in lipopolysaccharide-treated C3H/He mice. Concentration of serum amyloid substance in the serum of mice was lower in CCA-treated group than in the control group. CCA increased survival percent of mice injected with lipopolysaccharide. Histopathological investigation is now undergoing. Thus, CCA's biological activities so far clarified including the above-mentioned results in lipopolysaccharide-induced murine amyloidosis is characterized by its exclusive action in immuno-deficient hosts.
