<I>TEI-3096: An immunopharmacological profile</I>

TEI-3096 (6-<I>p</I>-chlorobenzyl-5H-2, 3, 6, 7-tetrahydro-5, 7-dioxothiazolo (3, 2-a) pyrimidine), a novel thiazolopyrimidine compound has been shown to suppress adjuvant arthritis in rats without any effect on conventional inflammation. We examined the immunopharmacological profile of TEI-3096 in murine lymphocytes. The blastformation induced by Con A or LPS was inhibited by addition of 5×10<SUP>-5</SUP>-5×10<SUP>-4</SUP> M concentrations of TEI-3096. This compound suppressed plaque-forming cell (PFC) response against sheep red blood cells (SRBC), a thymus-dependent antigen, irrespective of the concentrations of 2-mercaptoethanol (2-ME) in the culture medium. The suppressive effect was observed only when the drug was added to the medium within 48 hr after culture initiation. TEI-3096, however, produced either enhancement or suppression of PFC response to DNP-Ficoll, a thymus-independent antigen, depending on the dose of 2-ME. Repeated administration of TEI-3096 by oral route showed no significant effects on anti-SRBC PFC response in normal mice. However, this compound restored the colchicine-induced enhancement of antibody response against TNP-HGG. These results suggested that TEI-3096 might be interesting in that enhanced immune responses caused by immunological disorders might be normalized and might be useful for the treatment of autoimmune disease such as rheumatoid arthritis.
