Urinary excretion of prostaglandin E<SUB>2</SUB> and F<SUB>2α</SUB> was determined in rats by radioimmunoassay in order to study <I>in viva</I> inhibition of prostaglandin biosynthesis by non-steroidal anti-inflammatory drugs. The drugs were administered orally and the 4-hour urine was collected. Urinary prostaglandin E<SUB>2</SUB> as well as F<SUB>2α</SUB> decreased on indomethacin and mefenamic acid, and the decrease in these prostaglandins corresponded well with <I>in vivo</I> anti-inflammatory activity determined in rat carrageenin edema.
