Development of Bovine In Vitro Matured and Fertilized Oocytes Co-Cultured with Oviductal Epithelial Tissue in Serum-Free Medium

Examination was made of the effects of supplementation of fetal calf serum (FCS) on the blastocyst development of bovine in vitro matured and fertilized (IVM/IVF) oocytes in co-culture with bovine oviductal epithelial tissue (BOET). IVM/IVF oocytes were co-cultured in tissue culture medium 199 supplemented with or without FCS and/or BOET. The frequency of cleavage was the same with or without FCS (10%, v/v) and/or BOET (P>0.05). Blastocyst development by co-culture with BOET was higher than without BOET (P<0.001). When embryos were co-cultured with BOET, blastocyst development without FCS was similar to that with FCS (P>0.05). The developmental stage and total cell number of the blastocysts from co-culture without FCS were essentially the same as with FCS (P>0.05). Blastocysts from serum-free co-culture developed into normal calves after transfer.
