Fractionation and characterization of hydroxyethyl starch (I) which is used as a clinical plasma expander were studied. I was produced by the reaction of acid-hydrolyzed waxy corn starch with ethylene oxide. Fractionation was carried out by the fractional precipitation method in which I was precipitated by the addition of isopropanol to the aqueous solution. Solution properties of I such as viscosity, osmotic pressure, and light scattering were measured in various solvents. The results of the fractionation and the molecular weight measurements showed that the molecular weight distribution of I is extremely broad. The Mark-Houwink relationships between the intrinsic viscosities and the molecular weights were established. The exponents of the Mark-Houwink equation were 0.24-0.33. This result showed the feature of a highly branched polymer. On the basis of the above results, the effects of the solution properties of I on the biological activity as a clinical plasma expander were discussed.
