Ecdysteroid secretion in the nm-g mutant of the silkworm, whose development is arrested at the 1st or 2nd larval instar, was investigated. 1)The activity of the prothoracicotropic hormone (PTTH) in the brain of the mutant was high as that of a normal larva. 2)Injection of PTTH, bombyxin or brain extract into the mutant larvae did not induce molting. 3)Secretion of ecdysteroids by the prothoracic gland of the mutant was very low in vitro and was not activated by PTTH, bombyxin or the brain extract. 4) Ecdysteroid content in the prothoracic gland of the mutant was almost equal to that in the normal larva. 5)Ecdysteroid titer in the whole body of the mutant was low and constant for 5 days after ecdysis as in the case of the hemolymph. 6)Determination of ecdysteroid levels by HPLC-RIA showed that the mutant hemolymph contained small amounts of ecdysone and 20-hydroxyecdysone. On the basis of these results, we conclude that the abnormality in the nm-g mutant is caused by the low level of ecdysteroid synthesis by the prothoracic gland.
