In order to study the ecology of the white root rot pathogen of mulberry trees, the authors have developed a new method for isolating the pathogen from soil. The procedure is as follows: Small pieces of mulberry branch (2-3mm in diam., about 2cm in length) are mixed with the test soil containing the pathogen, and kept at 25°C for 5 days. Then, they are removed and placed on a medium suitable to identity the white root rot pathogen (PSA 1l+Anilin blue 0.2%+Nobobiocin 0.025%). Although many microorganisms appear from the pieces on the medium within a few days at 25°C, the pathogen can be distinguished because it specifically change the color of the medium to yellow.This method can be used to isolate the white root rot pathogen not only from the test soil in the laboratory, but also from the soil of mulberry field. Moreover, the rate of isolation is in proportion to the degree of contamination of soil with the pathogen.
