Enzymatic Histochemistry of the Rat Liver in Experimental Biliary Obstruction

Histopathological and histochemical studies have been carried out in she experimental biliary obstruction.<BR>1) Histopathological alterations showed a proliferation of bile duct epithelia with fibroblasts, mitotic figure of bile duct epithelia and hepatic cells, nodular hyperplasia of hepatic cells and disorganization of the liver pattern.<BR>There were only a few necrotic foci.<BR>2) Histochemical distribution of hydrolytic and oxidative enzymes, i. e., alkaline phosphatase, acid phosphatase, non-specific esterase, aminopeptidase, β-glucuronidase, succinate dehydrogenase, NAD dependent dehydrogenase (lactate, malate, glutamate, α-glycerophosphate, β-hydroxybutyrate) NADP dependent dehydrogenase (glucose-6-phosphate, isocitrate) and monoamine oxidase, were studied in the treated and normal livers.<BR>3) Histochemical alterations after biliary obstruction were observed during 6 weeks.<BR>Alkaline phosphatase and acid phosphatase activities increased in the treated hepatic cells and portal tracts, β-glucuronidase reaction increased in portal tract and non-specific esterase and aminopeptidase activity showed no alterations in them.<BR>Succinate, lactete, glutamate, β-hydroxybutyrate and glucose-6-phosphate dehydrogenase activities decreased in the periportal area and malate, α-glycerophosphate, isocitrate dehydrogenase and monoamine oxidase activities revealed no remarkable changes during the experiment.
