Effect of chamaecyparis obtusa oil on hepatic cytochrome P-450 isozymes in rat.

It has been known that <I>Chamaecyparis obtusa</I> (Hinoki Cypress), which is a useful Japanese tree, has various biological activities. To investigate the effect of <I>Chamaecyparis obtusa</I> oil on hepatic P-450 isozymes, we measured 1) testosterone hydoxylation activity and 7-ethoxycoumarin deethylation activity, 2) HPLC profiles of P-450, and 3) immunochemical quantitation of P450 PB-4 (P-450b), PB-5 (P-450e) and MC-5 (P-450c) by Western blotting in rat microsomes. As a result it was found that 1) an increase (50%, P<0.01) in hepatic P-450 content was caused by <I>Chamaecyparis obtusa</I> oil (300mg/kg, i.p., 2 times), 2) an increase (12-fold) in the content of P450 PB-4 and PB-5, which were strongly induced by Phenobarbital treatment, was shown, 3) the induction of P450 PB-4 and PB-5 was also supported by the measurement of testosterone hydroxylation activity and HPLC profiles, and 4) P-450 MC-5 was not induced, and no increase was also clarified by the measurement of 7-ethoxycoumarin deethylation activity. Therefore, we concluded that hepatic P-450 isozymes, PB-4 and PB-5, were induced by the administration of <I>Chamaecyparis obtusa</I> oil in rats.
