To evaluate the grade of malignancy in thymoma epithelial cell, we examined the aberrant expression of p53 in thymic epithelial tumor tissues immunohistochemically using polyclonal antibody CM-1. In addition we examined the expression of the proliferating-cell nuclear antigen to clarify the relationship of the expression of p53. The results of staining for p53 expression in stage, I, II and III thymomas in formalin-fixed specimens were all negative, but three stage N thymomas, two metastatic lung lesions, and two pleural dissemination lesions were positive. The result of staining for p53 expression of stage I thymomas in frozen specimens were all negative, but two stage III thymomas, two stage N thymomas, one metastatic lung lesion, and two pleural dissemination lesions were positive. On the other hand, concerning PCNA expression of thymomas, almost all epithelial cells and some lymphocytes were positive, and there was no relationship between the number of positive cells and the clinical stage of thymomas. Judging from these results, it was suggested that the expression of p53 might be a marker of the degree of malignancy in thymomas.
