CHARACTERIZATION OF CELL GROWTH INHIBITOR FROM BOVINE CEREBRAL CORTEX

Many trophic factors have previously been reported. However, few authors have reported on growth inhibitory factors, such as cell growth inhibitory factors purified from conditioned cell culture media or sialoglycopeptide obtained by protease E treatment of cerebral cortex homogenates.<BR>For the elucidation of PC-12 cell growth inhibitory factor, trypsinization, heat treatment and ion-exchange column chromatography were carried out. The cell growth inhibitory activity was found in the CM-TOYOPEARL bound fraction. One μg of protein from the above fraction inhibited PC-12 cell growth about 30% and the inhibition was reversible. Trypsinization was necessary to obtain the inhibitory protein.<BR>Our inhibitory protein specifically inhibits the growth of PC-12 cells. The growth of Neuro2 a and A375 cells was not inhibited. This inhibition spectrum is quite different from that of sialoglycopeptide, which inhibited the growth of both PC-12 and Neuro2a cells.
