With the object to study the course of healing of surgical wound and the effect of various drugs on it, wounds in rabbits were pathohistologically observed for 24 hours to 5 months after operation with the following results : <BR>Drugs used in the present experiments were the following six, all of which were locally applied to sutured site.<BR>1. A 10, 000-fold nitrogen mustard solution, intradermally injected in a dose of 0. 1cc.<BR>2. About 0.1g of 1% hydrocortisone ointment, applied once daily for 10 consecutive days.<BR>3. A 10-fold dilution of Typhogelison, intradermally injected in a dose of 0. 1cc.<BR>4. About 0.1g of Hirudoid ointment applied once daily for 10 consecutive days.<BR>5. A 10, 000-fold Quinhydrone solution (containing 3% carboxymethylcellulose) externally applied.<BR>6. A 5, 000-fold Quinhydrone solution, externally applied.<BR>Results were as follows : <BR>1. In the healing course of sutured wouhd of the rabbit, restoration of the epidermis preceded that of the connective tissue, and regenerated epidermis turned internally, iuvading the wound.<BR>The restoration of connective tissue was performed mainly by the upward proliferation of lower reticular layer of the corium, and muscular layer, which play a main role in fibroplasia. In the reticular layer, all tissue responses were weak through the whole course of healing.<BR>Elastic fibers showed a sign of regeneration in 3 to 4 days, and on the 7th day, obvions regeneration was observed in myoblastic tissue.<BR>There was no evident increase in PAS-positive substance, and it was scarce in regenerated collagenons fibers.<BR>2. After the intradeimal injection of 0.1cc of a 10, 000-foId nitrogen mustard solntion, and of 0.1cc of a 10-fold Typhogelison solution, and external application of a 10, 000-fold Quinhydrone solution, exudative response was not elicited in any case. And the formation of myoblasts was slight, fibroplasia rapid, without accompaniment of the degeneration of fibroblasts, regeneration of epidermis accelerated, and shrinkage of cicatrix effected.<BR>After external application of a 1% hydrocortisone ointment and a 5, 000-fold Quinhydrone solution, there were degeneration and destruction of fibrils, shrinkage of their nuclei, poor formation of myoblasts accompanied by vacuolation, and delayed fibroplasia, indicating imperfect healing. These treatments are therefore ineffective for surgical wound.<BR>The application of Hirudoid ointment produced remarkable delay in fibroplasia, and has unsatisfactory effect, when used immediately after operation.<BR>The effect of these drugs on elastic fibers and P AS-positive substance was not so remarkable. And the regeneration of elastic fibers tended to delay and PAS-positive substance tended to decrease in proportion with the degree of tissue injury.
