Previous stud : es have demonstrated that osteomyelitic staphylococci readily provoke multiple abscesses in long bones of young rabbits to a significant degree compared to other strains and produce coagulase most abundantly at pH. 8.3-7.7 in 90% of these strains, whereas other strains do so at pH. 6.7-7 0. In this papers it has been studied as followings : <BR>1). strains purely cultivated from 14 cases of acute osteomyelitis produce coagulase with optimum at pH. 8.0-7.7, including exceptional 2 strains from which such sub-cultures have been selectively obtained, and 16 strains cultivated from the other infections and 2 strains selected from the subcultures of the osteomyelitic 2 strains do so at pH. 7.4-7.0. The isoelectric point of the coagulases originated from osteomyelitic strains is at pH. 2.8-2. 5 culculated by acid precipitations distinctly from the others being at pH. 4.0-3.8. So purified coagulasea have been harvested most abundantly by the aids of these optimal production and isoelectric point, showing 672 to 465 fold purer than the crude materials as culculated on the basis of activities to a mg. of protain concentrations.<BR>2). Purified coagulases originated from osteomyelitic strains and the other strains are clearly differentiated in each others by zone electrophoresis with starch as supporting media, and especially separated in phosphate buffer at pH. 6. 8. The electrophoretically separated coagulases have shown 2 to 3 fold purer than the ones by acid precipitations.<BR>3). Anticoagulases recovered homologously, to osteomyelitic coagulases or the others in each immunitized rabbits. The patients of osteomyelitis and the other infections, although small cases, have shown specific immunological responces in the neutralisation of the coagulases originated from osteomyelitic strains and the others.<BR>In 1948 Tager found two mobil components in electrophoresis of coagulases, in 1950 Rammelkamp discovered three types of coagulases which produced specific anticoagulase and in 1958 Barber found specific anticoagulase related to phage groups, but had no attentions to the differences in the infectious diseases.
