Involvement of p21/waf-1 in Differentiation of Oral Epithelium in vivo and in vitro

The purpose of the present study was to investigate the expression pattern of p21/waf-1 in oral epithelium in vivo and in vitro. Immunofluorescence and in situ hybridization studies were performed to observe the expression of p21/waf-1 in tongue epithelium of rats. To induce differentiation of epithelial cells in vitro, epithelial rests of Malassez were cultured in a low Ca2+ medium, and then the medium was changed to a high Ca2+ medium. p21/waf-1 mRNA levels were analyzed by Northern blot. In vivo p21/waf-1 expression was observed in the upper spinous and granular layers of the epithelium. In vitro expression of p21/waf-1 mRNA was up-regulated after exposure to high Ca2+ medium. These results suggested that p21/waf-1 is involved in the differentiation of oral epithelium.
