Antihypertensive Activities of Beni-Koji Extracts and .GAMMA.-Aminobutyric Acid in Spontaneously Hypertensive Rats.

Beni-koji was extracted with ethanol and fractionated with ethyl acetate, n-butanol or water, in order to elucidate its antihypertensive activity. These fractions were fed to spontaneously hypertensive rats (SHR) in 1% NaCl diets for 15 days. The blood pressure of the group fed water-extract lowered. In another line of experiment, SHR were fed diets supplemented with γ-aminobutyric acid (GABA), which is a hypotensive component of the water-extract of beni-koji (0.09mg/100g diet). Although the control diet raised the blood pressure of SHR by 10mmHg, 0.1mg GABA/100g diet did not. However, addition of 1mg GABA/100g diet significantly repressed it similarly to the water-extract fraction. I. v. injection of water-extract fraction and 1mg GABA/rat induced the hypotensive effect in SHR.These results indicate that GABA is one of the important antihypertensive substances in beni-koji and the others are soluble in water and ethanol.
