A study was conducted to determine the effects of dietary icosapentaenoic acid (IPA) on plasma lipid metabolism. Twelve women (mean age of 22.3 years) participated. After one week on a control diet without IPA, an experimental diet enriched with sardine (IPA daily intake: 3.9g) was given for another 2 weeks with (IPA group) or without (Fish group) additional IPA capsules containing 1.8g/day. The following results were obtained.1) The serum IPA concentration increased from 17.5±4.2 (SE) to 195.0±2.4μg/ml in the Fish group and to 277±14.3μg/ml in the IPA group.2) Serum triglyceride levels tended to decrease and apolipoprotein C II concentrations significantly decreased in both groups.3) The triglyceride content of the very-low-density lipoprotein (VLDL) fraction decreased in both groups, and the decrease was larger in the IPA group.4) The cholesterol content of the low-density lipoprotein (LDL) fraction slightly reduced in both groups. Additional IPA supplementation significantly decreased the serum concentrations of total cholesterol and apolipoprotein B, associated with a decrease in the apolipoprotein content of the LDL fraction. These results suggest that dietary IPA supplementation decrease VLDL synthesis at a daily intake of 3.9g, produces a concomitant decrease in LDL synthesis at a higher intake amounting to 5.7g/day, and that the supplementation may be favorable for preventing vascular atheroscierotic changes.
