In order to clarify the function of nicotinic acid, changes in tyrosine hydroxylase activity in the brain of rats fed a tryptophan-imbalanced diet were studied.1) Tyrosine hydroxylase activity in the brain of rats fed a tryptophan-imbalanced diet was twice as high as that of rats fed a nicotinic acid-supplemented diet. The concentration of nicotinic acid in the brain and liver of the tryptophan-imbalanced rats was lower than that of the nicotinic acid-supplemented rats.2) The concentration of nicotinic acid in the brain and liver, and the activity of tyrosine hydroxylase in the brain returned to normal after the administration of nicotinic acid or a tryptophan-supplemented diet for 6 days to the rats fed a tryptophan-imbalanced diet.3) The concentration of nicotinic acid in the brain and liver was the highest 4 hours after intraperitoneal injection of nicotinic acid into rats fed a tryptophan-imbalanced diet. Tyrosine hydroxylase activity in the brain decreased to the normal level within 24 hours after the intraperitoneal injection.4) No remarkable change in tyrosine hydroxylase activity was observed by the addition of nicotinic acid or pyridine nucleotides to the brain homogenate of the tryptophan-imbalance rats.
