H89, a Protein Kinase Inhibitor, Promotes the Development of Thermotolerance in BHK21 and tsAF8 Cells.

The effects of Calphostin C (CAL ; a potent inhibitor of protein kinase C (PKC)), and H89, a protease inhibitor that inhibits c-AMP dependent protein kinase (PKA) and c-GMP dependent protein kinase (PKG), on thermotolerance of BHK21 and tsAF8 cells were investigated in this study. The cells were preheated at 45°C for 20 min and incubated at 37°C (BHK21) or 34°C (tsAF8) for 6 h with CAL or H89. They were then heated at 45°C and the surviving fractions were examined. The thermotolerance of the cells was suppressed by 2.5-10μM CAL (BHK21) or 0.25-0.5μM CAL (tsAF8), but was promoted by 2.5 or 5.0μM H89. When the cells were treated with H89 (0.025-30μM) between two heatings of 45°C for 20 min and 80 min (BHK21) or 40 min (tsAF8), the surviving fractions of both of the cell lines became the relative minimum at a concentration of 0.05μM, and the maximum at 2.5μM. The concentrations of these minimal and maximal points were close to those of H89 which inhibited PKA and PKG respectively. In conclusion, our results suggest that thermotolerance of BHK21 and tsAF8 cells may be developed by PKC and PKA but suppressed by PKG.
