Evaluation of Antibody-producing Activity and Occurrence of Antibody-dependent Cell-mediated Cytotoxicity(ADCC) in Chick Embryos.

Occurrence and presence of antibodies in the peripheral blood, IgM antibody-producing cells (IgM-PFC) in the spleens and activity of antibody-dependent cell-mediated cytotoxicity (ADCC) in the peripheral blood were examined in 15th to 20th embryonic-day-old chick embryos using xenogeneic red blood cells as antigens. Antibodies against goat red blood cells (GRBC) in the peripheral blood were detected just after hatching but could not be detected during embryonic development in the individuals immunized in embryonic period. IgM-PFCs producing anti-GRBC antibody in the spleens were detected for the first time just before or after hatching. These results indicate that antibody and IgM-PFC against GRBC were appeared at roughly equal stage of development. Presence of cells involved in ADCC against duck red blood cells were supposed to be already present in the peripheral blood of 15th embryonic-day-old chick embryos, and ADCC activity elevated during the embryonic days following. These results suggest that ADCC may contribure in non-specific elimination of foreign material, and, a small quantity of antibody and FC receptor-positive cells involved in ADCC may already be present in the peripheral blood of embryos during the late stage of embryonic development in chickens.
