Electrophoretic studies on carcinoplacental alkaline phosphatase (Nagao)

We have encountered a patient with gastric carcinoma metastatic to the liver, who had heat stable alkaline phosphatase in his serum and body fluid. The properties of his carcinoplacental alkaline phosphatase were studied and found almost identical to Nagao isoenzyme.However, this carcinoplacental alkaline phosphatase showed a broad isozyme band from the position of variant alkaline phosphatase to placental alkaline phosphatase on a thin layer polyacrylamide gel electrophoresis of 5-7% gel concentration. It was separated into two parts, fast and slow, from a 6% polyacrylamide gel slab and the properties of them were studied.The data obtained was almost identical to each other, although some microheterogeneities existed. The isozyme bands of this carcinoplacental alkaline phosphatase were also electrophoresed as the fastest migrating isozyme band, when polyacrylamide gel of over than 8% concentration was used.It is suggested that the epigenetic modification may cause microheterogeneities of this isozyme except for the effect of genetic expression of molecular structure.
