About 5.2 percent of endothelial cells covering not only unaffected parts, but also atheroma of cholesterol-fed rabbits, exhibited a strong contraction with pinches. Such contraction and blebbing, accompanied of the stretching of endothelial lining on the elevated plaque by the pulsation, are interpreted as the key mechanism in the progression of atheromatous lesions. The administration of estrogen, pyridinolcarbamate or EG467 exhibited a definite relaxing effect on the endothelial cells covering atheroma, and this fact suggests an explanations for the demonstrable efficacy of these agents ; endothelial-cell relaxants (Shimamoto 1972<SUB>2)-5)</SUB>), in preventing, arresting or regressing atheroma.
