Relationship between the Nutritional Status and the Alkaline Phosphatase Activity of the Serum (Report 1):On the Method of Measurement of the Serum Alkaline Phosphatase

Serum alkaline phosphatase activity was determined in blood of 20 adults, twenty-eight to seventy-three years old, and 20 children, aged six to eighteen, also of 20 rats, sixty to seventy days old, by the method of Nakagawa, using sodium paranitrophenyl phosphate as the substrate.Released para-nitrophenol was measured spectrophotometricaly using a Hitachi electric spectrophotometer at 415mμ and a Hitachi photoelectric calorimeter at 430mμ. As the optical dencities of paranitrophenol at 430mμ are lower than at 415mμ, so the standard courve are equivalent to 10mM unit at 415mμ but also are little equivalent to 8mM unit at 430mμ.For adult sera, which have low phosphatase activity, 0.1cc of serum, 1cc of reagent and 10cc of 0.02N NaOH were used at 415mμ, but the volumes of serum may be doubled, 0.2cc at 430mμ.For children sera, volumes of serum, reagent and 0.02N NaOH are increased to 0.1cc, 1cc and 20cc respectively.For rat sera, 0.05cc of serum, 1cc of reagent and 20cc of 0.02N NaOH were used.From the above mentioned procedure, as determined at 430mμ, is equivalent to 1.08±0.10mM unit in adult sera, 1.09±0.11mM unit in children sera and 1.04±0.07mM unit in rat sera at 415mμ.
