A Study of Lymphocyte Subsets in Patients Treated with Regional Hyperthermia for Advanced Urological Malignancies

Using FITC conjugated monoclonal antibodies (OKT3, OKT4, OKT8 and B1) and flow cytometry, peripheral blood lymphocyte subsets were examined before and after 8 MHz-RF hyperthermia in 33 patients with advanced urological malignancies. Hyperthermia was performed alone or in combination with irradiation (radiohyperthermia) or anticancer drugs (chemohyperthermia). Eleven patients with benign diseases were similarly evaluated to compare with the patients with malignancies. Between the patients before hyperthermia and the patients with benign diseases there were no significant differences in the percentages of OKT3<SUP>+</SUP>, OKT4<SUP>+</SUP>, OKT8<SUP>+</SUP> and B1<SUP>+</SUP> lymphocytes respectively. In the radiohyperthermia and chemohyperthermia groups, the total blood lymphocyte counts after treatment were significantly reduced, however, those of hyperthermia alone group showed no significant change. Statistically significant increases in the percesntages of OKT3<SUP>+</SUP> and OKT8<SUP>+</SUP> lymphocytes were found in the group of patients who showed no response after radiohyperthermia. No groups had remarkable changes in the percentages of OKT4<SUP>+</SUP> and B1<SUP>+</SUP> lymphcytes and the OKT4<SUP>+</SUP>/OKT8<SUP>+</SUP> ratios. It seemed that local hyperthermia by itself may not affect the total blood lymphocyte counts and the proportions of these lymphocyte subsets except for the combination with irradiation or anticancer drugs.
