We examined the reaction surfaces around two variables (lactate and NAD<SUP>+</SUP>) in the JSCC (Japan Society of Clinical Chemistry) method for determining Lactate dehydrogenase (LD), using 23 factorial experimentation at the reaction temperatures of 30 and 37°. Theoretical response surfaces were computed by fitting a quadratic polynomial equation to the experimental data by least-squares regression. Our response surface data showed a broad plateau over the concentration range investigated (lactate 5-210 mmol/L, NAD<SUP>+</SUP> 1-19 mmol/L, respectively ). Response surface methodology (RSM) is a collection of statistical and mathematical techniques useful for developing, improving and optimizing processes.
