EFFECTS OF N-(ETHYL-2-PYRROLIDINYL-METHYL)-2-METHOXY-5-SULFAMOYL BENZAMIDE (SULPIRIDE) UPON THE GASTRO-INTESTINAL MOV EMENTS

1) In dogs anesthetized with pentobarbital sodium, intravenous injection of sulpiride (0.05-10 mg/kg) produced in most cases an excitatory effect, occasionally an excitatory effect followed by an inhibitory one and in rare cases only an inhibitory effect in the gastrointestinal movements. After cutting the extraintestinal nerves the effects described above remained the same.<BR>2) The drug, in the concentration of 10<SUP>-5</SUP>-5x10<SUP>-4</SUP> g/ml, produced the augmentation or inhibition in the motility of the isolated intestine of dogs, rabbits and rats.<BR>3) After administering tetrodotoxin or atropine the excitatory effect of the drug was abolished, whereas the inhibitory one remained the same.<BR>4) The contraction of frog rectus abdominis induced by acetylcholine (10<SUP>-7</SUP>g/ml) was increased in its height by applying sulpiride (10<SUP>-6</SUP>-5x10<SUP>-4</SUP> g/ml).<BR>5) The excitatory effect of the stimulation of the vagus nerve upon the gastrointestinal movements was increased by administering sulpiride (2-10 mg/kg.)<BR>6) From the results described above it may be concluded that sulpiride, on the one hand, stimulates the intramural excitatory, cholinergic neurones to enhance the release of acetylcholine, which is responsible for augmenting the gastrointestinal motility, and on the other hand, exerts a depressant action upon the gastrointestinal muscle. In addition, the evidence has been obtained that the drug suppresses the action of acetylcholinesterase.
