The effects of glycyrrhizin on the pituitary adrenal axis were studied. According to the method of Wool <I>et al</I>, the isolated anterior pituitary glands of the rats were incubated with <SUP>14</SUP>C-phenylalanine and the rates of <SUP>14</SUP>C-corticotropin and <SUP>14</SUP>C-protein synthesis in the pituitaries were measured. In agreement with the results of Wool <I>et al</I>, it was found that the rate of <SUP>14</SUP>C-corticotropin synthesis was increased in the pituitaries of the adrenalectomized rats, while adrenalectomy did not increase the incorporation of amino acid into the total protein of anterior pituitary. <BR>Identification of the extracted radioactivity with corticotropin was performed on a horizontal starch block electrophoresis, and it was found that the radioactivity consisted of two fractions electrophoretically, each of which had high activity of corticotropin. <BR>The rate of <SUP>14</SUP>C-corticotropin synthesis was depressed below normal in the pituitaries of prednisolone treated rats. Cortisone treatment suppressed the increase of <SUP>14</SUP>C-corticotropin synthesis induced by prior adrenalectomy. However, concomitant injections of glycyrrhizin blocked the inhibitory action of cortisone on <SUP>14</SUP>C-corticotropin synthesis. Injections of glycyrrhizin alone had no effect on the <SUP>14</SUP>C-corticotropin synthesis in the pituitaries of both normal and adrenalectomized rats. <BR>Compensatory adrenal hypertrophy of rats after unilateral adrenalectomy was completely suppressed by cortisone treatment, and the concomitant injections of glycyrrhizin partially but significantly blocked the suppression of compensatory adrenal hypertrophy by cortisone treatments. Glycyrrhizin alone had no effect on the compensatory adrenal hypertrophy. <BR>From clinical studies, it was also recognized that glycyrrhizin effectively prevented the suppression of the pituitary adrenal axis by glucocorticoid. Namely, oral administration of glycyrrhizin in daily doses of 150-300 mg blocked the effect of 1-2 mg of dexamethasone in lowering the amout of total urinary 17-OHCS as the result of pituitary inhibition. <BR>The depletion of adrenal ascorbic acid contents of the rats by histamine stress was examined after cortisone treatments. Simultaneous injections of glycyrrhizin increased the inhibitory action of cortisone on adrenal ascorbic acid depletion under the condition of stress. <BR>The possibility of dissociation of the secretory mechanism of corticotropin in the basal or resting stage from that of the condition of stress is being discussed, and the mechanism of the ihibition of glycyrrhizin on the action of cortisone and possilble clinical use of glycyrrhizin are also being discussed.
