The present investigation was undertaken to see whether or not any correlation exists between morphological changes of the adrenal cortex of rats and production of aldosterone and corticosterone from the gland under various conditions : injection of cortisone, ACTH or DOCA, and feeding with sodium deficient diet. Production of aldosterone and corticosterone was determined according to the procedure of Saffran and Shally, and paper chromatography of Bush B<SUB>5</SUB> system. <BR>The experimental results obtained from this work are summarized as follows : <BR>1) <I>Sodium deficient diet</I>. It was observed in rats fed with sodium deficient diet for 10 consecutive days that the width of zona glomerulosa was thickened, their cells enlarged, and that the sudanophil droplets became fine. An increase of aldosterone production was also confirmed in this group. <BR>2) DOCA <I>injection</I>. An increased production of both aldosterone and corticosterone was noted in the adrenal slice of the rats injected with 200 μg/d of DOCA for 10 days. It was found that the width of zona glomerulosa was slightly reduced in this group. <BR>3) <I>ACTH injection</I>. In rats injected with ACTH 2 units daily for 10 days, aldosterone was produced in an increased amount while zona glomerulosa nearly disappeared. On the other hand, it was found that zona fasciculata was hypertrophied, and corticosterone production was increased. <BR>4) <I>Cortisone injection</I>. Rats treated with cortisone (2.5 mg/d × 10 d.) showed an increase of the width of zona glomerulosa. In contrast to sodium deficient diet group, sudanophil granules did not become finer under cortisone treatment. <BR>On the basis of these data, it seems reasonable to assume that morphological findings of zona glomerulosa of the adrenal cortex could not be regarded as an indicator of the production of aldosterone when any alteration of zona fasciculata is detected as seen in cases of ACTH or cortisone. It could be supposed from the results of the ACTH group that aldosterone might be produced not only from zona glomerulosa but also from the other zones. Conversely, the morphological findings of zona fasciculata were found to parallel the production of corticosterone from the adrenal cortex.
