When Hizikia fusiformis chloroplasts were solubilized by a solubilization buffer prepared at a weight ratio for SDS: Triton X-100: chlorophyll (Chl) of 20:10:1, the solubilizing ratio was 82.8%, but it was 31.9% when using a buffer containing only Triton X-100 at a Triton X-100/Chl weight ratio of 25:1. When chloroplasts were solubilized by a buffer prepared at a ratio of 10:20:1 and subjected to SDS-PAGE, the area ratio of brown-orange zone IV to the free pigment (FP) zone was 0.283. In the isolation of CPC from H. fusiformis by SDS-PAGE with the Tris-borate fuffer (Tris-B) system, the migrating degree of the FP zone became high in proportion to an increase in the elec-trophoretic time as compared with the migrating degree of CPC zones. In SDS-PAGE with the Tris-B system, the migrating speed of the FP zone was exceedingly high in comparison with that in the Tris-HCl buffer (Tris-H) system, and the migrating degrees of CPC (zones I-IV) were smaller than those in the Tris-H system. By using the electrophoretic procedure with the Tris-B system the time could be shortened and the amount of FP could be reduced in comparison to that with the Tris-H system.
