Furosemide enhances gentamicin induced inhibition of Na<SUP>+</SUP>-dependent D-glucose transport in renal proximal brush border membrane vesicles

Furosemide is known to potentiate gentamicin nephrotoxicity. We studied the effects of gentamicin (G) and/or furosemide (F) on Na<SUP>+</SUP> -dependent D -glucose transport into brush border membrane vesicles (BBMV) isolated from rabbit kidney outer cortex (OC : early proximal tubule) and outer medulla (OM : late proximal tubule). Rabbits received either 1 ml of normal saline (group-C), G (20 mg/kg) alone (group-G), F (10 mg/kg) alone (group-F) or combination of G and F (group-GF) intravenously. After 1 hour, kidneys were removed and BBMVs were then prepared by the calcium precipitation method. Kinetic analyses revealed that G alone impaired only the D -glucose transport into OC-BBMV, increasing K<SUB>m</SUB> value from 3.5±0.3 nm/min mg prot to 6.3±0.2 (p<0.05). When F was co-administered, D-glucose transport was impaired both in OC and OM BBMVs. F alone had no significant effect on either OC or OM BBMV D-glucose transport. There was no difference in the osmotically active space and the passive permeability among the BBMV groups. F may potentiate G toxicity on BBMV D-glucose transport by enhancing GM accumulation (74.4±12.4μg/g wet tissue, G alone vs 112.7±26.9, G+F, in OC and 82.5±18.3 vs 153.5±37.4 in OM).
