A lectin is a protein being able to bind with certain structure of sugar chains. Therefore, many lectins have been used as tools to detect various sugar chains on cell surface membranes. Though there are a number of methods for this purpose, i. e., agglutination test and microscopic or electron microscopic observation of cells treated with fluorescence or ferritin labeled lectins, this article deals with lectin binding assay which gives quantitative data on the cell surface sugar chains. The number of specific sugar chains (lectin receptors) and the association constant (<I>K</I>a) between the lectin and the sugar moiety are obtained by this method. Furthermore, the equations to calculate the inhibition constant (<I>K</I>i) of unlabeled lectin or inhibitory sugars are also introduced. The actual applications of these methods are also mentioned, i. e., the number of blood group O determinant on human red blood cells was calculated by the binding assay with eel serum anti-H (O) agglutinin and some sugar structures of the lectin receptors on human red blood cells were estimated by comparison of <I>K</I>a values and <I>K</I>i values.
