The mechanism of riboflavin-mediated photo-generation of superoxide anion (O2_??_) in milk serum was studied employing model systems con-taining milk serum proteins or amino acids and riboflavin. Among various systems tested, those containing β-lactoglobulin, α-lactalbumin, or cysteine were effective in generating O2_??_. The photogeneration of O2_??_ in the cysteine-system was completely eliminated by treatment of cysteine with iodoacetic acid. Reduced glutathione (-SH) but not oxidized glutathione(-S-S-) was also effec-tive in photogeneration. These results suggest that SH groups play an important role in the riboflavin-mediated photogeneration of O2_??_ as an electron donor to photo-excited riboflavin.
