The distribution of free sugars in the seeds, leaf buds, leaves, stalks and others of various vegetables (including thirty-one genera and ten families) during the course of germination of seeds and in the ripening periods has now been investigated by the use of paper partition chromatography. The developing solvent systems used were (a) n-butanol: acetic acid water (4:1:2 by volume), and (b) n-butanol: pyridine: water (3:2:1.5 by volume). Chromogenic spray reagents used were (a) an ethanolic solution of benzidine-trichloroacetic acid, and (b) an ethanolic solution of phloroglucinol-trichloroacetic acid. The multiple ascent technique was used for the method of development of chromatograms of mixed sugars. Chromatograms of mixed sugars were usually run simultaneously on the same sheet of paper with those of the mixture of authentic specimens. Raffinose has been shown to be co-existent with sucrose in the seeds of all the vegetables examined in this investigation. Stachyose has been shown to be co-existent with sucrose and raffinose in the seeds of twenty-four kinds of various vegetables. Sucrose has been shown to be co-existent with glucose in the leaf buds of all the vegetables examined in this investigation. Fructose has been shown to be co-existent with sucrose and glucose in the leaf buds of eleven kinds of various vegetables.
