1. Amount of arginine in the hydrochloric acid-hydrolyzates of bull semen, spermatozoa, and seminal fluid was estimated colorimetrically by a method based on the Sakaguchis reaction. 2. In four experiments water contents of spermatozoa, and seminal fluid were found to be 83.45 and 90.70 per cent, respectively. The weight of one spermatozoon was determined to be 8.32×10-5γ. (3) The analytical method was shown to be sensitive to 15.8_??_58.0 micrograms of arginine in 1.5_??_2.9mg. of the fresh samples of spermatozoa. (4) The average values of arginine in spermatozoa were 2.05 per cent on the fresh basis, and 13.32 per cent on the dry basis. (5) Arginine was ascertained in the fresh sample of seminal fluid as 0.33 per cent on the wet basis, and 3.79 per cent on the dry basis.
