A specific radioimmunoassay (RIA) was developed for measuring lactoferrin (LF) in aspirate by using the pancreozymin-secretin (PS) test. The method utilized the double antibody RIA technique using radioiodinated LF purified from human colostrum, rabbit antibody to LF, and sheep antibody to rabbit IgG. The minimal sensitivity of this RIA method was 2ng•er milliliter. The mean concentration of LE was significantly higher in chronic pancreatitis than in the control, about thirty five times higher at the pancreozymin phase, thirty two times higher at the secretin phase in chronic calcifying pancreatitis, thirteen times higher at the pancreozymin phase, and five times higher at the secretin phase in non-calcifying pancretitis.When abnormal level of LF was shown as 476ng/ml=(Mean+2SD)×2 in the control group, abnormality was observed in 6 of 10 patients with calcifying pancreatitis (60%) and in 4 of 11 patients with non-calcifying pancreatitis (36%), but not in 8 patients with pancreatic cancer.The measurement of LF in aspirate by PS test using the RIA method can be an effective diagnostic method in chronic pancreatitis.
