Recently, prostaglandin (PG) cytoprotection has been of great interest in the gastrointestinal tract, but there are few reports about endogenous PGs in gastric mucosa. We established a method of determining intramucosal PGE2 in the previous study. We estimated here the levels of B-, E1-, E2-, F-type of PG and we have investigated the effect of stress on PGE2 and cyclic AMP levels in rat gastric mucosa.The level of intramucosal PGE2 was markedly high compared with other types of PG both in fundus and in pyloric antrum in the untreated group. In the stress series, the PGE2 levels was increased in the early phase in which any mucosal lesion was not observed. Then it was decreased in the late phase accompanied with production of the gastric mucosal lesion. The cyclic AMP level was also increased in the early stress phase while it was not influenced with stress when pretreated with indomethacin.These results suggest that the lack of intramucosal PGE2 may cause production of the stress ulcer.
