Hepatic cell necrosis in rabbits was experimentally produced by administration of lymphokine either intraperitoneally or into the hepatic portal vein which appeared in culture supernatants of stimulated lymphocytes obtained from rabbits immunized with rat liver homogenates. The severity of the necrosis was roughly proportional to the dose of the lymphokine and submassive necrosis was observed in the most severely affected animals. The lymphokine showed dose-dependent cytotoxicities against isolated rat hepatocytes and Chang liver cells in vitro, but not against murine L929 cells. The molecular weights were between 40, 000 and 90, 000 according to Sephadex G-100 gel filtration. The cytotoxic activity was disappeared following heating at 56°Cfor 30 minutes or by trypsin digestion, but was not affected by neuraminidase treatment. These results suggested that a hepatocytotoxic lymphokine played an important role in the progression of hepatic necrosis and that this lymphokine was different from lymphotoxin.
