Effects of endothelin-1 on the contraction of Ito cells (fat-storing cells).

In the present study, to examine whether Ito cells are able to contract, the binding of endothelin-1 (ET-1) on cultured Ito cells and changes of their areas induced by ET-1 were investigated by autoradiography using <SUP>125</SUP>I-ET-1 and addition of ET-1 to the culture medium. A remarkable accumulation of <SUP>125</SUP>I-ET-1 grains was observed on the surface of Ito cells. The area of Ito cells gradually decreased following ET-1 stimulation from 4×10<SUP>-10</SUP>M to 8×10<SUP>-7</SUP>M in a dose dependent manner. These results may indicate that Ito cells are contracted by ET-1 via ET-1 receptors. Fathermore, in this study, it is suggested that Ito cells may be related to the regulation of hepatic sinusoidal microcirculation through the contraction of their long cytoplasmic projections.
