We studied the metabolic response of ketone body to glucose load in rat liver with and without acute CCl<SUB>4</SUB> intoxication. The metabolic changes after oral glucose load were as follows:<BR>1) α-glycerophosphate concentration remained unchanged in CCl<SUB>4</SUB> liver, whereas it rose to 1.7-fold above the initial level in control liver.<BR>2) Oxaloacetate concentration dropped to about 50% below the initial level in CCl<SUB>4</SUB> liver mitochondria, whereas it rose to 3-fold above the initial level in control liver mitochondria.<BR>3) β-hydroxybutyrate/acetoacetate ratio remained statistically unchanged in CCl<SUB>4</SUB>liver, suggesting little suppression of the rate of oxidation of fatty acids in CCl<SUB>4</SUB> liver mitochondria.<BR>4) As the result of these evidences, acetyl-CoA concentration of CCl<SUB>4</SUB> liver remained almost unchanged resulting in a little lowering of ketone body concentration in CCl<SUB>4</SUB> liver.
