Crystal structures for two [2Fe-2S] ferredoxins as well as that of a [4 Fe-4S] ferredoxin from <I>Bacillus thermoproteolyticus</I> were determined at 2.5 Å resolution. Predicted tertiary structures of various [2 Fe-2S] ferredoxins and [4 Fe-4S] ferredoxins of sulfate reducing bacteria were given in the present study. Tertiary structures of main-chains and substitution frequency along the polypeptide chain for the [2Fe- 2S] ferredoxins show that the less important in physiological functions a residue is, the less conservative during the evolution. The reversion mutation proposed by Yanofsky <I>et al</I>. ( Science, 148, 1593-1594, (1964) ) was demonstrated in tertiary structures of [2 Fe- 2S] ferredoxins. These evolutional phenomena are consistent with the nutral theory of molecular evolution. Evolution from the [8Fe- 8S] ferredoxin to the [4Fe- 4S] ferredoxins are proposed by tertiary structural similarity of these ferredoxins.
